Default Nuts and washer on inner tube? Use or Lose?

RV7A build

Does anyone know the purpose of the Nut and Washer at the base of the inner tube stem closest to the tube?

The inner tube comes with 1 nut and 1 washer at the base of the stem and 1 nut beneath the valve stem cap. I have read in the Matco instructions that nut/washer at the base of the stem stays on and should be placed in the slot on the rim when mounting.
Would anyone know how tight the nut at the base of stem should be when installing?

As for the nut at the end of the stem near the valve stem cap I need to know If and How I should use that.

I have Grove wheels and Michelin tubes. More hardware than places to put it.
